Lecture 3 Lab Design, Layout & Equipment
  • 6 main functions
    • sterile handling→end of room, far from door
    • incubation & storage→readily accessible to work area
    • preparation→adjacent to washup & sterilization
    • washup & sterilization→at other end
  • layout depends on types & scale of operation
    • user number
    • space→4 culture operation: 2 washup, preparation, sterilization: storage: incubation
    • location of prep area
    • storage
    • access
    • biohazard protection & sterility
      • containment requires contents of tissue culture room not escape to adjacent work areas
      • asepsis requires none of contamination of surrounding areas enter tissue culture
  • Equipment
    • sterile handling→BSC, laminar flow, pipette, inverted microscope, centrifuge, cell counter
    • incubation: CO2 incubator
    • prep: washup facilities, water purifier, balancer, magnetic stirrer, pH meter
